摘    要:为研究有利于菜心出芽的生长素-细胞分裂素(NAA-BAP)浓度配比,以‘油青’菜心的带柄-子叶为外植体,在MS培养基中分别加入不同浓度配比的生长素和细胞分裂素,共设置16个处理观察菜心带子叶的子叶柄出芽情况。试验结果表明,不同浓度配比NAA-BAP对菜心出芽影响不同,菜心出芽量在各处理间有显著差异。当培养基中NAA-BAP浓度配比为0.5~1 mg/L、0.25~2 mg/L时有较高的出芽数量和出芽率,表明此浓度配比最适合菜心出芽。这为菜心的转基因研究、快速繁殖等提供了基础。

Abstract:

In order to determine the mixing concentration of auxin and cytokinin (NAA-BAP) which was conducive to the budding of Brassica campestris ssp. chinensis Makino var. parachinensis (Bailey) Tsen et Lee to help further agrobacterium-mediated transformation. This test uses MS medium added with mixing concentration of auxin-cytokinin has been used for cultureing cotyledon petioles of Brassica campestris ssp. chinensis Makino var. parachinensis (Bailey) Tsen et Lee, and also observed the budding. The results showed that mixing concentration of auxin-cytokinin have different effects on cotyledon petioles of Brassica campestris ssp. chinensis Makino var. parachinensis (Bailey) Tsen et Lee budding, when the mixing concentration of auxin-cytokinin for 0.5-1 mg/L, 0.25-2 mg/L were suitable for Brassica campestris ssp. chinensis Makino var. parachinensis (Bailey) Tsen et Lee budding. So cotyledon petioles of Brassica campestris ssp. chinensis Makino var. parachinensis (Bailey) Tsen et Lee budding affected by hormones also needs more extensive research.
