乌菜组织培养及耐热变异体的诱导和筛选

摘    要:以安徽地方品种“合肥黄心乌”的下胚轴、子叶、茎尖组织为外植体,在无菌条件下接种在不同植物激素的MS培养基上。在适宜的温度和光照培养条件下,获得大量的下胚轴组织形成的愈伤组织,其出愈率为75.5%,愈伤组织转入MS+8mg/L6BA的培养基上,分化出不定芽,但分化率很低,仅为0.5%。茎尖组织在适宜的培养基上,直接产生芽簇。这些不定芽和芽簇经切割继代培养,形成正常植株。在乌菜组织培养快速繁殖的基础上,进行耐热诱导和定向筛选。经不同温度和不同时间的处理,有10.6%的小苗再生植株,这些植株移入大田后,在延后栽培的高温下能正常生长,但影响结实率,仅有30%左右的植株正常结实。再生植株后代的耐热稳定性及品质在进一步试验之中。

Abstract:Cultured on MS medium with hormones for 25 days,hypocotyl and cotyledon of Wucai formed callus,but frequency of callus was 75. 5%,and stem tip straightly formed bud cluster. Callus cultured on MS medium cantaining singBAL-1 could differentiate into plantlets and differentiation frequency was only 0. 5%. 10. 6 per cent of planls could survivethrugh high temperature for the plantiets and bud cluster. These heat-resistant plants could mormally grow at 35 ℃,buttheir fruiting rates were lower. Only 30% of plants were fertile. Heat-resistant' stability and quality of generation plantprogeny are being studied.
