扬州鹅PRL基因内含子2的SNP检测及其与产蛋性状的相关分析

摘    要:
为提高扬州鹅的产蛋性能,采用PCR-SSCP方法检测催乳素基因内含子2上的SNP位点,并分析其与扬州鹅产蛋性状的相关性。结果表明:扬州鹅PRL基因内含子2上存在1个SNP位点,以内含子2的起点开始算起,在第32、33位点上发生了GA碱基的插入/缺失,导致其产生3种基因型。等位基因A、B的频率分别为0.586 3、0.413 7;基因型AA、AB、BB的频率分别为0.461 9、0.248 7、0.289 4。分析表明:这3种基因型的分布不符合哈代温伯格平衡,内含子2的突变位点对扬州鹅的产蛋量和开产日龄都没有显著的基因型效应,但在产蛋量和开产日龄上都有BB型>AA型>AB型的趋势。

Abstract:
PRL is an important candidate gene,which influences the avian species'broodiness and egg performance.The egg performance of geese is low and has breeding seasonality.Therefore,in order to increase their egg performance,the single nucleotide polymorphism(SNP) sites of PRL gene intron 2 were detected in Yangzhou geese by PCR-SSCP method and the correlation analysis between the polymorphism of PRL gene intron 2 and egg-laying traits was carried out.The results showed that there is a mutation in intron 2 of PRL gene,GA insertion/deletion at the site of 32 and 33 nucleotides,which resulted in 3 genotypes(AA,BB and AB).The result showed that allele frequency of A and B are 0.5863 and 0.4137,and the genotype frequency of AA,BB and AB are 0.4619,0.2487 and 0.2894,respectively.Analysis showed that the distribution of the three genotypes was inconsistent with the Hardy-Weinberg equilibrium.Mutation of intron 2 of Yangzhou geese have no significant effect on the egg production and the age at first egg,but in egg production and the age at first egg of genotype are BB>AA>AB.
