昆明裂腹鱼两地理群体遗传多样性的随机扩增多态性DNA(RAPD)分析

摘    要:应用随机扩增DNA多态性(RAPD)技术对昆明裂腹鱼盘大河群体和昆明裂腹鱼泥猪河群体的遗传多样性进行分析。从38个10bp引物中选取20个用于群体遗传多样性分析,在两群体中分别检测到121、125个位点,多态位点数分别为57、73个,平均多态位点比例分别为47.11%、58.40%;两群体内个体间平均遗传距离分别为0.1745、0.1963,群体间遗传距离为0.2503。研究表明:因地理隔离的两昆明裂腹鱼群体间存在一定的遗传分化;两群体内和群体间的遗传变异较大,具有丰富的遗传结构;泥猪河群体的遗传多样性要比盘大河群体的遗传多样性丰富,其群体内遗传变异较盘大河群体大。

Abstract:Random amp lified polymorphic DNA (RAPD) technique was employed to analyze the genetic diversity two stocks of Schizothorax Grahami in Pan Da-he river and Ni Du Zhu-he river. Using 20 out of 38 decamer random primers ,the RAPD loci of two stocks were 121 and 125,of which 57 and 73 loci were polymorphic. The stock of polymorphic loci of two stocks was 47.11% and 58.40%.The average genetic distances among individuals of two stocks were 0.174 5 and 0.196 3.
