水稻原生质体高效培养技术的研究

摘    要:
用16个基因型水稻成熟胚在MS和N6培养基上筛选建成悬浮细胞系9个,原生质体再生植株3个。使用一步法或两步法较常规的分步法缩短建成悬浮细胞系时间20~30天。使用“三合一”培养基(即N6大量元素、MS微量元素和B5有机物等)和固液双相培养法较琼脂糖包埋法及液体浅层培养法显著提高原生质体培养的植板率。

Abstract:
Nine suspension cell lines were established from mature embryos of sixteen genotypes of rice in MS and N6 medium.Three genotypes of protoplast plants were regenerated.Instead of traditional method,"one step method"or"two step method"shortened 220-30days in the estabfishmen of suspension cell lines. Compared with agrose imbed culture method and thin liquid culture method,"three in one"medium (i. e. N6 macroelement,MS microelement and B5 organic compound etc.)and solid plus liquid culture method obviously increased protoplast culture efficiency.
