摘    要:本文介绍了用 PEG 和蔗糖密度梯度区带离心纯化猪瘟病毒的方法和步骤。先用 PEG 沉淀浓缩猪瘟兔化弱毒感染的牛睾丸细胞培养液,再经蔗糖密度梯度区带离心,出现4条区带,其中病毒感染力最强的位于Ⅱ带,其次为Ⅰ带,蛋白质含量多为0.14~0.19毫克/毫升和0.21~0.12毫克/毫升。电镜检查可见带有膜囊的病毒粒子,SDS—聚丙烯酰胺凝胶电泳显示Ⅰ、Ⅱ、Ⅲ、Ⅳ带均各出现一条明显蓝色条带外,在它前后侧还隐约可见一条浅带。用这些方法浓缩提纯兔化猪瘟弱毒效果较好,且较简便易行。

Abstract:The lapinized Chinese strain of hog cholera virus cultivated in fetal testis mo-nolayer culture was concentrated by means of polyethylene glycol(PEG)preci-pitation.The concentrated virus was then again purified by zonal centrifugation insucrose density gradient.Electron microscopic examination and SDS-PAGE of thefinal preparation revealed that the lapinized Chinese strain of hog cholera virusparticles had been highly purified.
