摘    要:以霍霍巴为材料 ,研究了外植体的消毒、基本培养基、取材部位和培养条件等因素对组培苗生长的影响。结果表明 :采用 0 .1 %HgCl2 与 2 %NaClO 1∶1混合的消毒合剂处理外植体 8min ,灭菌成功率 99% ;改良MS培养基适合霍霍巴的分化培养 ;最适外植体为双腋芽茎段 ;培养条件 :温度 2 5~ 2 8℃ ,光照强度 30 0 0lx ,光照时间 1 2~ 1 4h/d ,pH 5 .5~ 6.0。

Abstract:The key techniques including explants disinfection, basic medium, suitable explant and culture condition were studied.By culturing the stems of Jojoba in vitro,the results showed as that the success percentage of disinfection reached 99% when the explants were treated by 1∶1 mixture of 0.1% HgCl 2 and 2% NaClO for eight minutes,the modified MS was suitable for Jojoba micropropagation.The optimal explants were stems with double axillary buds.All the cultures were kept under the temperature of 25-28 ℃,light intensity 3000 lx,light time for 12-14 h/d,and pH 5.5-6.0.
