| 摘 要： | 制备PrP(prion protein)多克隆抗体,验证原核表达的人PrP的抗原性,为PrP空间构象的转变机制等深层次的研究以及人PrP单克隆抗体的制备提供重要的试验材料.以融合的人成熟PrP蛋白(mature prion protein,mPrP)为抗原,以pET30a(+)空载体的E.coli BL21 (DE3)的菌体蛋白作为对照免疫原,免疫小鼠.ELISA和Western blot分别检测多克隆抗体的效价和特异性.结果5只动物中3只均产生了较高效价的抗血清,确定人 PrP多克隆抗体效价为1∶4 096,能与人mPrP特异性结合.证明获得的人mPrP具有良好的免疫原性.
