摘    要:
[目的]研究1株多粘类芽孢杆菌所产菊粉酶的粗酶学性质。[方法]以菊芋菊粉为底物发酵培养了1株多粘类芽孢杆菌,利用超声波法破碎细胞,得到该菌的菊粉酶粗提液,并对菊粉酶的粗酶学性质进行了研究。[结果]这株多粘类芽孢杆菌最佳产酶时间约在摇瓶发酵培养40h时,所产的胞内菊粉酶的菊粉酶活力与蔗糖酶活力之比(I/S)为0.2187,小于10,表现为外切酶活性。该酶反应的最适温度和最适pH值分别为45℃和5.0,在此条件下菊粉酶酶活力最高,可达102.81U/L。[结论]这株多粘类芽孢杆菌所产菊粉酶为外切酶,该酶的粗酶学性质研究为进一步深入系统地研究菊粉酶的酶学性质奠定了基础。

Abstract:
[Objective] The enzymatic property of inulinase from one strain of Bacillus polymyxa Mace was studied.[Method] One strain of Bacillus polymyxa Mace was cultured,the cells of the strain were broken with ultrasonic wave method and the crude extracts of the inulinase from the bacteria were obtained.The enzymatic property of the crude inulinase was researched.[Results] The best enzyme production of the strain was at about the 40th hour when it was cultured in shake flask,at that moment,the rate (I/S)of the acti...
