多叶型和三叶型苜蓿杂交F1代遗传关系的SSR分析

摘    要:为了分析苜蓿(Medicago sativa)多叶性状的遗传规律,以及多叶苜蓿和三叶苜蓿杂交F1代性状分离特性和群体结构特点,本试验以淮阴苜蓿和多叶苜蓿品系PL34HQ为亲本,人工杂交获得F1群体。对F1群体180株苜蓿材料进行农艺性状和经济性状的差异分析,并从分子水平上进行遗传多样性和群体结构的分析。结果表明,F1群体春季多叶率达到91.11%,单株多叶率最高为93.33%,最低1.64%,平均为39.8%,春秋季节的群体多叶率高于夏冬季节;F1群体中紫色茎、绿色茎和紫绿色茎的植株出现的比例近乎1:1:1,株型的分离比接近9:7:2。SSR标记共检测到79个等位变异,平均每对引物检测到4.16个,19对引物的平均多态性比率是61.58%。群体结构分析将F1分为4个类群,类群2的遗传多样性最高,其次是类群1和类群4,类群3的遗传多样性水平最低。

Abstract:To elucidate the separation characteristics and the genetic characteristics of multifoliolate traits of F1 hybrid generations of trifoliolate alfalfa and muitifoliolate alfalfa, Huaiyin alfalfa and muitifoliolate alfalfa PL34HQ were used to hybridize to generate reciprocal crossing F1 generation plants. 180 plants of F1 generation were used to analyze agronomic and economic characters, genetic diversity and population structure. Results showed that:the group multifoliolate rate of F1 population was 91.11%, the maximum multifoliolate rate of the plants with multifoliolate trait was 93.33%, and the minimum was 1.64%, 39.8% was the average. Group multifoliolate rate of F1 population in spring and autumn was higher than summer and winter. The proportion of purple stems, green stem and purple-green stem was nearly 1:1:1, and the proportion of plant type separated rate was about 9:7:2. A total of 79 allelic variations were detected with SSR markers, and 4.16 to per primer, the average ratio of polymorphism was 61.58%. F1 population was divided into four groups by genetic structure analysis, and the results of genetic diversity of the cluster analysis showed that highest genetic diversity existed in Group 2, followed by Group 1 and Group 4. Genetic diversity levels of Group 3 was the lowest.
