Abstract:Sapium sebiferum(Linn.) Roxb.is most important of woody oil tree species in China,but the related SRAP-PCR application on it has been hardly reported.Based on the template DNA of superior tree of Sapium sebiferum(Linn.) Roxb.,the optimum SRAP-PCR reaction system for Sapium sebiferum(Linn.) Roxb.was established as follows: 2μL10×PCR Buffer,0.2mmol/L dNTPs,0.25mmol/L primer,1.5U Taq polymerase and 40ng template DNA,the total reaction volume was 20μl.And 27 primer combinations which produced clear bands and with abundant polymorphism were screened from 100 combinations.The optimized SRAP-PCR reaction system and polymorphic primer combinations provide some technical foundation for the further related genetic analysis of Sapium sebiferum(Linn.) Roxb.Resoures.
