刺芹侧耳多肽对染铅大鼠脑海马组织形态及海马组织中camk2a、erk2和homer1基因mRNA表达量的影响

摘    要:为研究刺芹侧耳(Pleurotus eryngii)多肽对染铅大鼠脑组织损伤的影响,以雄性SD大鼠为研究对象,随机分为正常对照组(基础饲料,蒸馏水)、染铅模型组(基础饲料,0.2%醋酸铅水溶液)和多肽组(添加2%多肽的基础饲料,0.2%醋酸铅水溶液),饲养60d后,观察大鼠脑部海马组织形态变化,测定海马组织中铅含量、camk2a、erk2和homer1mRNA的表达量。结果表明:铅会导致海马组织的细胞数量减少、细胞层变薄、细胞间隙增大、camk2a、erk2和homer1mRNA表达量极显著降低;与染铅模型组相比,多肽组海马组织的铅含量显著降低、细胞数目增多、细胞核变大、细胞间隙缩小,同时海马组织中NMDAR和mGluR5信号通路相关基因camk2a、erk2和homer1mRNA的表达量显著或极显著升高,表明刺芹侧耳多肽可减轻铅对大鼠海马组织的损伤。

Abstract:To exlpore the effect of Pleurotus eryngii polypeptides on brain injury in lead acetate-exposed rats.Male SD rats were randomly divided intothree groups:control group (basal feed, distilled water), lead-exposed model group (basal feed, 0.2% aqueous solution of lead acetate) and polypeptide group (basal feed supplemented with 2% polypeptide, 0.2% aqueous solution of lead acetate).After 60 days of polypeptide feeding, the results showed lead acetate could decrease number of cells in hippocampus, thickness of cell layer, expression levels of camk2a, erk2 and homer1 mRNA, and increase cell gap in lead acetate-exposed model control group.Compared with the lead-exposed model group, the polypeptide treatment could significantly induce decrease in lead content, increase in number of hippocampus cells, larger cell nucleus, and narrower cell gaps as well as significantly higher or extremely significantly higher expression levels of camk2a, erk2 and homer1 mRNA relating to NMDAR and mGluR5 signaling pathways in hippocampus.These results indicated that P.eryngii could relieve the impairment of rat hippocampus induced by lead acetate.
