8种产地铁皮石斛蛋白质和同工酶分析

摘    要:研究可溶性蛋白和同工酶电泳图谱与铁皮石斛种质资源之间的关系。利用可溶性蛋白和同工酶电泳技术,对来自8个产地的8种铁皮石斛的可溶性蛋白、过氧化物酶(POD)、多酚氧化酶(PPO)和超氧化物歧化酶(SOD)进行聚丙烯酰胺凝胶电泳分析。电泳分析表明,8种铁皮石斛材料共得到119条谱带,特异性条带6条,多态性谱带占总谱带数的94.6%。在3种同工酶中,POD的谱带最为丰富,8种材料共得到36个条带;而PPO的谱带最少,8种材料只得到22个条带,说明其蛋白质和同工酶在种类和表达量上均有比较明显的差异。对8种铁皮石斛电泳条带进行的主成分分析表明,8种产地铁皮石斛的遗传变异不同,可分为4类。该研究对铁皮石斛的引种、育种具有指导意义。

Abstract:The purpose was to study the relationship between soluble protein and isoenzyme electrophoretogram and germplasm resources of D.officinale.The soluble protein and isoenzyme electrophoretic techniques were used to analyze the soluble protein,POD,PPO and SOD in 8 materials of D.officinale from 8 producing areas with the polyacrylamde gel electrophoresis.The result showed that total of 119 bands were obtained from 8 D.officinale materials including 6 specific bands and 94.6% of the total bands were polymorphism bands.Among 3 kinds of isoenzyme,the spectrum bands of POD were richest and total of 36 bands were obtained from 8 kinds of materials;the spectrum bands of PPO were least and only total of 22 bands were obtained from 8 kinds of materials,indicating that their proteins and isoenzymes both had more obvious differences in the species and expression quantity.The principle component analysis of the electrophoretic bands amplified from 8 kinds of D.officinale showed that the genetic variations of D.officinale from 8 producing areas were different and could be classed into 4 categories.The research had guiding significance in the introduction and breeding of D.officinale.
