西瓜高效再生体系的建立

摘    要:以优质小果型西瓜品种西173和小麒麟为材料,系统研究了以子叶为外植体的西瓜组培高效再生体系。结果表明:2个西瓜品种在不定芽和愈伤组织诱导上存在着较大差异,在MS+6-BA1.0mg·L-1+IBA0.5mg·L-1的分化培养基上,西173不定芽诱导率最高达到92.6%,在MS+6-BA2.0mg·L-1+IAA0.2mg·L-1培养基上小麒麟不定芽诱导率最高达到89.7%;诱导出的不定芽丛伸长以MS+KT0.2mg·L-1培养基最佳;2个西瓜品种不定芽在MS+IBA0.2mg·L-1培养基上均获得了较好的生根效果。组培苗移栽后保持高温、高湿是移栽成活的重要条件。西瓜高效再生体系的建立为应用基因工程技术改良西瓜品质和提高其抗逆、抗病性奠定了基础。

Abstract:A high frequency watermelon regeneration system had been developed by using watermelon cultivar X173 and Xiaoqilin.The results showed that MS+6-BA 1.0 mg·L-1+IBA 0.5 mg·L-1 was the most suitable medium for bud organogenesis from cotyledon explants of X173 and MS+6-BA 2.0 mg·L-1+IAA 0.2 mg·L-1 for Xiaoqilin,with regeneration frequency of 92.6% and 89.7%,respectively.The suitable shoot elongation medium for both cultivars was MS+KT 0.2 mg·L-1.The favorable rooting medium for both cultivars was MS + IBA 0.2 mg·L-1.Warm and humid condition is necessary for plantlets to survive after transplant into soil.Our regeneration system should be helpful watermelon genetic transformation and genetic improvement via in vitro technology.
