粘液繁殖体植物抱茎独行菜TTG1基因转化拟南芥的表型研究

摘    要:目的]将过量表达典型粘液繁殖体植物抱茎独行菜TTG1基因(LpTTG1)的转基因拟南芥株系表型与野生型和突变体进行比较,为深入研究LpTTG1基因的功能提供参考依据.方法]在体视显微镜下,通过对拟南芥野生型、ttg1突变体及LpTTG1基因过表达转基因株系的植株表型,幼苗叶表皮毛、下胚轴和根毛以及种子形态进行观察比较.结果]拟南芥ttg1突变体与野生型和LpTTG1过表达转基因型的幼苗在下胚轴的颜色、根毛着生方式、真叶表皮毛等方面有显著差异,但后两者表型间无显著差异.结论]缺失TTG1基因可导致拟南芥幼苗形态发生显著变化,但此基因过量表达不一定对拟南芥形态产生影响.

Abstract:【Objective】To study the phenotype change in transgenic Arabidopsis with LpTTGl gene(from Lepidium perfoliatum,a typical myxospermy species) overexpression,and in hope of being able to provide evidence for exploring the biological functions of LpTTGl.【Method】With stereomicroscope,the plant phenotype,trichome of young seedling leaf,hypocotyl color,root hairs and seed morphology were observed in wild -type,ttg1 mutant and overexpression LpTTGl gene Arabidopsis.【Result】The color of hypocotyl,the way of root hair growth and the leaf trichome presented obvious difference between ttgl mutant and wild - type or LpTTG1 overexpression Arabidopsis,however,no significant difference was observed between wild - type and LpTTG1 overexpression Arabidopsis.【Conclusion】Loss of TTG1 gene can cause apparent changes in morphology of Arabidopsis seedling,while overexpression of this gene is not likely to affect the morphology of Arabidopsis.
