Abstract: | AIM: To study the effects of β1,3-N-acetylglucosaminyltransferase 8 (β3Gn-T8) on the biological behaviors of gastric cancer cells and xenograft tumors. METHODS: After pEGFP-C1-β3Gn-T8 was transfected into human gastric adenocarcinoma AGS cells, the cell proliferation was measured by MTT assay and viable cell counting with trypan blue exclusion, and the apoptotic rate was analyzed by flow cytometry. The model of AGS cell subcutaneous xenograft was established to detect the growth of the xenograft. RESULTS: The proliferation of AGS cells transfected with pEGFP-C1-β3Gn-T8 was significantly increased (P < 0.05), but the apoptotic rate was not significantly changed. The growth rate and final volume of xenograft tumor in pEGFP-C1-β3Gn-T8 group were significantly higher than those in control group (P < 0.05). CONCLUSION: β3Gn-T8 promotes the proliferation of gastric cancer AGS cells and significantly increases the growth of AGS cell xenograft tumor. |
