金桔无性快繁体系建立的研究

摘    要:以金桔的成龄叶片、带腋芽茎段及种胚为外植体,将它们接种于附加不同激素的MS培养基中,并进一步诱导分化出芽及再生植株。结果表明:剥去种皮的种胚诱导效果远好于成龄叶及茎段的诱导效果。而MS+蔗糖30g/L且不添加生长调节剂的配方最适宜种胚初代诱导。诱导获得的上胚轴节段在MS+BA2.0mg/L+NAA0.5mg/L+蔗糖30g/L+琼脂7g/L的培养基上进一步分化成苗,将无根苗接在1/2MS+NAA0.02mg/L的培养基中,经3周培养生根。

Abstract:The adult leaf,stem segment with axillary bud,seed embryo of kumquat were used as explants,they were cultivated on MS medium containing different hormones for the inducement of sprouts and regenerated plantlets.The results showed that the inducement effect of the naked seed embryo was much better than that of adult leaf and stem segment.MS + 30 g/L sucrose without any growth regulators was the best culture medium for the initial inducement of seed embryo.The epicotyls explants obtained from seed embryo were cultivated on the culture medium MS + BA 2.0 mg/L + NAA 0.5 mg/L + sucrose 30 g/L + agar 7 g/L,and they were able to develop into plantlets.Then the plantlets could take roots on the medium 1/2MS + NAA 0.02 mg/L after 3 weeks'culture.
