摘    要:以卷丹百合(Lilium lancifolium)鳞茎鳞片为外植体,探讨影响其分化的主要因素。正交试验结果表明,诱导卷丹百合鳞茎鳞片不定芽的最佳培养基为MS+6-BA 2.0 mg/L+NAA 0.1 mg/L,诱导鳞茎鳞片愈伤组织最佳培养基为MS+6-BA 1.5 mg/L+NAA 0.2 mg/L;继代培养中发现,小鳞茎在添加50 g/L蔗糖MS培养基中生长量最大,添加40 g/L蔗糖有利于愈伤组织生长,且小鳞茎生根率高,生根数量多。

Abstract:The study used squamae of Lilium as explants to explore the key factor that affecting differentiation of them.The results of orthogonal designed test showed that the best formula for primary culture of the squamae scales was MS+6-BA 2.0 mg/L+NAA 0.1 mg/L.The best formula for primary culture of the squamae callus was MS+6-BA 1.5 mg/L+NAA 0.2 mg/L.During the secondary culture,small bulbs kept the largest growth in MS culture with 50% sugar concentration,which was good for callus growth with 40% sugar concentration,and small bulb had a high rooting rate and large quantity root.
