骨碎补水提液对大鼠成骨细胞OPG mRNA表达的影响

摘    要:通过探讨骨碎补水提液对大鼠成骨细胞中OPG mRNA水平表达的影响,来研究骨碎补对成骨细胞产生作用的分子机制。细胞培养48h后,应用荧光定量RT-PCR方法检测不同浓度骨碎补水提液及雌激素受体抑制剂ICI 182780作用后,细胞中OPG mRNA水平表达的变化。结果显示,骨碎补水提液作用于大鼠成骨细胞48h后,与对照组相比,高浓度骨碎补水提液显著上调OPG mRNA的比值,而低浓度组与对照组无显著差异。加入雌激素受体抑制剂ICI 182780后,骨碎补对OPG mRNA水平的促进作用消失。表明骨碎补水提液能上调OPG mRNA水平的表达,且呈剂量依赖性。上述促进作用能被雌激素受体抑制剂阻断,因而可以推测这个过程与雌激素受体通路有关。

Abstract:The objective of present study was to explore the effects of rhizoma drynariae on the mRNA expression of osteoprotegerin(OPG) in rat osteoblasts.The third-passage of cultured osteoblasts was exposed to different concentrations of the water extract of rhizoma drynariae for 48 h.The total RNA was extracted,and the change of OPG mRNA was observed by fluorescence quantitative real-time RT-PCR.The results showed that after 48 h treatment of rhizoma drynariae,at high concentrations,the water extract of rhizoma drynariae up-regulated significantly OPG mRNA,while at lower concentrations there were no effects.After adding the ER binding inhibitor,ICI-182,780,the effects of rhizoma drynariae on OPG production disappeared.These results demonstrate that the stimulation of OPG appear to be mediated,at least in part,via a genomic pathway operating through estrogen receptors and gene expression mechanisms in rat osteoblasts.
