转反义TrxS基因导入小麦后子粒发芽能力与脱支酶活性的变化

摘    要:对转反义TrxS基因的小麦株系(00 T 89)进行纯合株系的子粒发芽试验和脱支酶活性、可溶性糖含量测定.结果表明,在开花后35 d至成熟后10 d,00 T 89株系子粒发芽开始时间比对照推迟2 d,子粒发芽率和发芽指数分别比对照降低22.1%和35.4%,差异达到显著或极显著水平.转基因小麦子粒的脱支酶活性在成熟至成熟后10 d平均比对照降低了27.3%,达到显著水平(P<0.05);在开花后35 d至成熟后20 d,转基因子粒可溶性糖含量平均比对照降低了31.6%,达到极显著水平(P<0.01).子粒发芽特性与脱支酶活性及可溶性糖含量呈显著的相关关系.

Abstract:After transferring the anti-TrxS gene into wheat,the seeds germinating ability,the debranching enzyme activity and the soluable sugar content in the seeds of pure transgenic line 00 T 89 were studied.The results showed that the transgenic line had stronger resistance to seed germination.Compared with non-transgenic plant,the seeds germinating time in 00 T 89 was postponed by 2 d,and the seed germination ratio and the germination index were reduced by 22.1% and 35.4% respectively in the period of 35 d after anthesis to 10 d after ripeness.The difference was significant.The DBE activity in 00 T 89 was decreased by 27.3% significantly(P<0.05) 10 d after ripeness.The soluble sugar content was decreased significantly by 31.6% from 35 d after anthesis to 20 d after ripeness(P<0.01).Based on further analysis,the correlation among the seeds germinating ability,the debranching enzyme activity and the soluble sugar content reached a significant level.
