棉属种间杂交基因渐渗系SSR标记及其表型性状的聚类分析

摘    要:利用37对多态性SSR引物和15个表型性状对155份棉属种间杂交基因渐渗系及其10份陆地棉亲本进行了聚类分析。用37对SSR引物共扩增出108条带,多态性条带占85.2%。种质间成对相似系数平均值为0.6791,范围为0.4762~1.000。建立了165个陆地棉材料的SSR聚类图,共聚为4个类群,分别为法国种质群、美国AcalaSJ系列种质群、美国PD种

Abstract:The main cultivated varieties belonged to the species of Upland cotton(Gossypium hirsutum L.) in the world and their genetic background is very narrow.However,The wild species and race in the genus of Gossypium have abundant genetic polymorphisms and possess lots of excellent genes with potential high yield,fine and strength fiber,disease and insect resistance,drought and coldness resistance,male sterility and so on.The interspecific hybridization for utilizing the benefit genes from these wild species had been widely carried out for 15 years in China.It has huge practical and theoretical value to conduct the analyses of the cotton introgression lines of interspecific hybridization toward upland cotton,which help to increase the genetic basis of the present upland cotton resources and break the bottleneck of cotton breeding.In this paper,the cluster analysis of 155 introgressed lines from interspecific hybridization and 10 parents of Gossypium hirsutum was conducted by using 37 pairs of polymorphic SSR primers and 15 phenotype traits(Table 1).Based on 37 pairs of polymorphic SSR primers,108 bands altogether were amplified and the percent of polymorphic bands was 85.2%.Average paired similarity coefficient among germplasm was 0.6791,ranged from 0.4762 to 1.000.The SSR cluster dendrogram of 165 varieties of Gossypium hirsutum was constructed(Fig.2) with 4 groups: the French group,the American AcalaSJ series,the American PD lines and the germplasm in China.The result of SSR cluster analysis fitted basically with that of their pedigrees.The Euclidean distance was 0.641710.3075 among different germplasm with an average of 5.0047.The 155 introgressed lines were divided into 9 groups by cluster analysis of their phenotypes(Fig.3).The agronomic and economic characters in different groups were significantly different.There was an obvious difference between the cluster analysis of phenotype traits and the pedigrees of these germplasm.Comprehensive utilization of SSR and phenotype trait cluster analyses will promote the use of the introgressed lines from interspecific hybridization in cotton.
