摘    要:目的]建立有效的植物内源激素提取方法,并探讨利用反相高效液相色谱法同时测定骆驼刺根状茎分株处3种植物内源激素的色谱分析条件。方法]采用反相高效液相色谱法,对骆驼刺分株处根状茎中植物内源激素赤霉素(GA3)、玉米素(ZR)和生长素吲哚-3-乙酸(IAA)进行了分离和含量测定。结果]GA3、ZR和IAA的平均回收率为98.3%,90.3%和101.3%,说明该方法符合定量分析要求,且误差小。分离骆驼刺根状茎植物内源激素的高效液相色谱条件为:以流动相甲醇:0.75%乙酸为45∶55、流速0.7 ml/min、检测波长254 nm和柱温为25℃。结论]该研究初步建立了分离骆驼刺根状茎植物内源激素的高效液相色谱条件。

Separation and Content Determination of Three Kinds of Endogenous Hormones in Rhizome of Alhagi sparsifolia

Abstract:Objective] The research aimed to establish an effective extraction method of plant endogenous hormones and discuss the chromatographic conditions for determining three kinds of plant endogenous hormones in rhizome ramet of Alhagi sparsifolia simultaneously by reversed-phase high performance liquid chromatography(RP-HPLC).Method] Using RP-HPLC,plant endogenous hormones gibberellin(GA3),zeatin(ZR) and auxin indole-3-acetic acid(IAA) were separated from rhizome ramet of A.sparsifolia and their contents were determined.Result] Average recovery of GA3,ZR and IAA were 98.3%,90.3% and 101.3% respectively,which indicated that this method could meet the demands of quantitative analysis and had little error.The conditions for separating three kinds of plant endogenous hormones from the rhizome ramet of A.sparsifolia by RP-HPLC were as follows:using methanol-acetic acid aqueous solution(with the volume ratio of 45∶55) as the mobile phase,the flow-rate of 0.7 ml/min,the detection wavelength of 254 nm and the column temperature of 25 ℃.Conclusion] The research preliminarily established the high performance liquid chromatography conditions for separating plant endogenous hormones from the rhizome of A.sparsifolia.
