摘    要:采用SDS-PAGE电泳技术,比较分析了双向电泳中常用4种染色方法的优、缺点。结果表明:常规银染法灵敏度低;改进的双胺硝酸银银染法与质谱分析不兼容;考染方法-银染复合染色法的灵敏度稍低于双胺硝酸银银染法,但它的质谱兼容性最高;改进后的银染方法灵敏度高、背景清晰、与质谱兼容。因此后两种方法较广泛适用于蛋白质组学研究。

Abstract:In order to choose an appropriate visualization method for proteomics research,the advantages and disadvantages of four protein staining methods in common use in two-dimensional electrophoresis(2-DE) were compared and analyzed by using SDS-polyacrylamide gel electrophoresis(SDS-PAGE).The results showed that the sensitivity of conventional silver staining method was low;the improved silver staining method of diamine silver nitrate was not compatible with MS;the sensitivity of the complex silver staining method combined with Commassic brilliant blue was slightly lower than that of the improved silver staining method of diamine silver nitrate;the sensitivity of improved silver staining method was high,having a clear background,being compatible with MS.Therefore,the latter two methods were more widely used in proteomics research.
