椰乳在甘蔗组培快繁中作用的研究

摘    要:以种性优良但组培分化斟难的3个甘蔗品种FN95-1702、FN99-2308和FN99-20169的愈伤组织为材料,以2种常用的培养基为基本培养基,分别添加4种不同体积的椰乳,采用随机区组实验设计,进行分化阶段和生根阶段培养,并对移栽幼苗成活率进行了观察.结果表明:对于不同激素配方的培养基,在分化阶段添加椰乳后甘蔗愈伤组织的分化率和芽长势均明显提高,添加椰乳的适宜体积为10%~20%,其中,以15%为最佳添加体积,可使分化率提高100%.幼苗生根阶段,添加椰乳使幼苗生根的数量减少,对移栽成活率影响不明显.不同甘蔗品种适合的培养基略有差异,实验所选用的2种培养基与椰乳之间无交互作用.

Abstract:Coconut water(CW) improves callus and shoot formation in tissue culture of some species of plants. No report about effect of CW on sugarcane tissue culture is found documented. Three sugarcane varieties, FN95-1702, FN99-2308 and FN99-20169, which have desirable novel traits but are difficult in shoot formation under tissue culture were selected for this purpose. Two hormones and four volumes of CW were added into MS medium, and randomized block experiments were designed. Calli induced from shoot tip of the 3 sugarcane were cultured on 2 different MS media at 4 levels of CW at the shoot formation and rooting stages. Survival rates of rooted plants transplanted were also observed to determine potential effect of CW on the growth of the tissue cultured plants. The results indicated that CW significantly improved shoot formation of all the sugarcane varieties tested in media at the CW levels from 100 mL/L to 200 mL/L, of which addition of CW with 150 mL/L improved callus differentiation up to 100%. The addition of CW into the rooting media reduced the number of roots but had no obvious effect on survival rates of sugarcane plants transplanted. MS medium suitable for culture of sugarcane varieties varied but no interaction between the media and CW were observed in this experiment.
