江南桤木等根瘤内生菌的分离和回接

摘    要:从赤杨属(Aluns)江南桤木(Alnus traboculosa Hand)根瘤上分离到放线菌Frankia sp.F_3,从胡颓子属(Elaeagnus)厦门胡颓子(Elaeagnusoldhani Maxim)根瘤上分离到放线菌Frankia sp.FE4,蔓胡颓子(Eleaegnusglabra Thumb)根瘤上分离到放线菌Frankia sp.FE_3,均获得纯培养。 用光学显微镜和扫描电镜观察其形态特征,确认是属具有孢囊的放线菌。该菌慢生长型,在培养基内菌丝体发育好,无气生菌丝体。液体培养,在底部生长,属微好气性。孢囊形状不规则,囊内包着大量的不游动的孢囊孢子。菌丝分枝有隔,粗细不匀,革兰氏阳性。将分离菌F_3,FE_4,FE_3回接其宿主,感染结瘤,有固氮活性。

Abstract:The nodule actinomycetes, Frankia sp, F3, F. sp. FE4, F. sp. FE3, have been isolated from Alnus traboculosa, Elaeagnus oldhani and E. glabra respectively and their pure cultures were obtained. The isolates from A. traboculosa and E. oldhani were identified as new strains.These actinomycetes belong to slowngrowing types, They have well-develo-ped mycelia in the media but they do not have any aerial mycelium. The irr egular sporangia would produce a large number of nonmoving sporangiospores. The hyphae were thin, branched,septate,inconsisleutly coarse,and Gram's stain positive. when inoculated with the isolates of F3, FE4 and FE3, the hosts were infected and produced nodules with significant" nitrogen fixation.
