罗美沙星在鸡体内的药物动力学与生物利用度研究

摘    要:采用反向高效液相色谱法, 研究了罗美沙星在健康鸡体内的药动学与生物利用度。结果表明, 鸡静注罗美沙星后的ci- ti数据符合二室开放模型, 其主要动力学参数如下: tt/2α056±012h, t1/β769±079 h, Vd216±035 L/kg, C L B020±003 L/kg·h, A U C 5219±899 ug/m L·h。鸡内服、肌注罗美沙星c1- t1 数据, 均符合有吸收因素二室模型, 内服的主要动力学参数如下: t1/2ka118±014 h、t1/2α245±046 h、t1/2β1721±158 h、 A U C 3878±633 ug/m L·h、tmax316+ 023 h、 Cm ax206±043 ug/m L、 F7431% ; 肌注的主要动力学参数是: t1/2ka067±012 h、t1/2α130±023 h、t1/2β1152±102h、 A U C 4315±204 ug/m L·h、tm ax176±020 h、 Cmax460±ug/m L、 F8268% 。本文还就罗美沙星的临床应用进行了药动学评价。

Abstract:The pharmacokinetics and bioavailability of lomefloxacin in healthy broilers were studied following single intravenous, oral and intramuscular administration at 10 mg/kg dosing rate by using a reversed phase high performance chromatagraphy (HPIC) procedure. The results obtained showed that the plasma c i-t i data of lomefloxacin after intravenous administration in healthy broilers were best described by a twocompartment open model. The main pharmacokinetic parameters were as follows:t 1/2α 0 56±0 12 h,t 1/β 7 69±0 79 h,Vd 2 16±0 35 L/kg,CL B 0 20±0 03 L/kg.h、AUC 52 19±8 99 ug·mL.h. The c i-t i data of lomefloxacin in plasma following both oral and intramuscular administration in healthy broilers were fitted to a two two-compartment open model with first-order absorption. The main parameters after oral administratiion were as follows:t 1/2ka 1 18±0 14 h、t 1/2α 2 45±0 46 h、t 1/2β 17 21±1 58 h、AUC 38 78±6 33 ug·mL.h、t max 3 16±0 23 h、 C max 2 06±0 43 ug/ml、 F74 31%;The main parameters after intramuscular administration were as follows:t 1/2ka 0 67±0 12 h、t 1/2α 1 30±0 23 h、t 1/2β 11 52±1 02 h、AUC 43 15±2 04 ug·mL.h、t max 1 76±0 20 h、C max 4 60±0 61 ug·mL、F 82 68%、The pharmacokinetic evaluation was alos conducted on the clinical applications of lomefloxacin in this report. $$$$
