国内外部分小麦种质资源高分子量麦谷蛋白亚基组成分析

摘    要:用十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)方法分析150份国内外小麦种质资源,并按照相关评分方法计算高分子量麦谷蛋白Glu-1位点的品质得分,结果表明:小麦种质资源的Glu-1位点具有丰富的遗传变异,共检测到15种亚基和31种亚基组合类型,品质评分为4~10分,平均为6.83分。在Glu-A1位点具有3种亚基类型(Null、1、2*),其中,null亚基频率高达58.7%;Glu-B1位点具有7种亚基类(6+8、7+8、7+9、13+16、14+15、17+18、7),7+9亚基类型出现的频率最高,为40.66%,其次7+8亚基频率高达37.3%;在Glu-D1位点上,具有5种亚基类型(2+12、5+10、4+12、3+12、5+12),2+12亚基出现频率最高达65.3%,5+10亚基出现频率为30%。具有较高品质得分的亚基组合"1、7+8、5+10"的品种有10份,优异种质资源可供优质小麦育种利用。

Abstract:The compositions of high molecular weight glutenin subunits(HMW-GS) of 150 wheat germplasms were analyzed using SDS-PAGE.The quality score at Glu-1 of each germplasm was calculated according to the quality rating system described by Payne et al.The results showed that the genetic variation at Glu-1 locus of introduced wheat germplasms was abundant,in which 15 subunit patterns and 31 subunit combinations were detected.The mean quality score of these germplasms was 6.83,ranging from 4 to 10.Among these 150 wheat varieties,subunit null was found in 58.7% Glu-A1 locus,subunit(7 9) and a allelic variation(7 8) were found in 40.66% and 37.3% Glu-B1 locus respectively.While at Glu-D1 locus,the frequency of subunit(2 12) was the highest of 65.3% and subunit(5 10) was 30%.Ten germplasms had the subunit combination 1,7 8 and 5 10 with higher quality score.And they could be widely utilized in wheat quality breeding.
