柴达木盆地梭梭CMO基因的克隆及其蛋白结构预测

摘    要:对柴达木盆地梭梭的CMO基因进行扩增,并利用生物软件对CMO基因序列进行分析、对蛋白结构进行预测。结果表明,柴达木盆地梭梭CMO基因长度为1 341 bp,编码447个氨基酸。CMO蛋白亲水性、二级结构、亚细胞定位、信号肽、跨膜螺旋区、三级结构预测结果显示柴达木盆地梭梭CMO蛋白亚细胞定位于叶绿体,无信号肽,蛋白二级结构主要为无规则卷曲。CMO蛋白具有多个多种类型的功能位点,为蛋白功能的实现提供了保障。此外CMO蛋白的亲水性较强,无跨膜螺旋区。柴达木盆地梭梭CMO蛋白三维结构预测结果显示CMO蛋白三维结构由α螺旋、β折叠和无规则卷曲互相盘绕而成。

Abstract:CMO gene of Haloxylon ammodendron ocurring in Qaidam Basin was amplified by PCR and it’s protein structure was predicted by biology software.The results showed that the length of the CMO gene and CMO protein were 1 677 bp and 559 aa respectively.The prediction results of hydrophilicity,secondary structure,subcellular localization,signal peptide,transbilayer helix and tertiary structure showed that CMO protein was located in chloroplast,without signal peptide,random coils were the main secondary structure of CMO protein and it had many kinds of functional sites.CMO protein had good hydrophilic property,and it had no transmembrane structures.The tertiary structure prediction showed the main tertiary structure of CMO protein included α-helix,β fold and random coil.
