摘    要:根据已发表的菠菜、烟草等植物GA 20-oxidase基因序列在保守区设计简并引物,通过RT-PCR和RACE的方法克隆了Marster香石竹(Dianthus caryophyllus L. cv. Marster)GA 20-oxidase基因的全长cDNA(1 179 bp),命名为Dc20ox。同源性分析表明该基因与其它作物上发表的GA 20-oxidase基因的氨基酸序列同源性为66%~75%。在此基础上选用同源性相对较高的400 bp DNA片段,构建了RNA干扰(RNAi)载体pART400。

Abstract:A pair of degenerate primers were designed based on the conserved domain of the GA 20-oxidase reported in other plants. A full-length cDNA (1 179 bp) of carnation (Dianthus caryophyllus L. cv. Marster) GA20-oxidase named Dc20ox was cloned by RT-PCR and RACE methods. The deduced amino acids of this sequence shared a high homology. Blast showed that the amino acids sequence presented a high homology (66%~75%) with the GA20-oxidase in other plants. A RNAi vector (pART400) was constructed by a high homology 400 bp fragment.
