Identification of a natural variant of African swine fever virus in China

African swine fever(ASF)has been introduced into China for more than two years since 2018,the genomes of African swine fever virus(ASFV)strains of Chinese origin submitted to the GenBank up to date has been genetically unchanged substantially.However,in view of the possible slowdown of ASF epidemic and continuously notified low mortality of African swine fever,we performed an ASFV ecology investigation,and isolated a natural variant of African swine fever virus from Hubei.In genome sequencing,a partial deletion,with a total 1252 bp,of EP402 R gene(CD2 v)and its upstream adjacent EP153 R gene(8 CR)was identified,resulting in loss of haemadsorption.A number of unique nucleotide mutations,deletions or insertions in several gene coding and intergenic regions occurred,which are different from all those of the 10 known ASFV strains in GenBank derived from China.Some of the variations lead to the changes of its coding amino acids and open reading frames(ORFs)and even disappearance of ORFs where the variations locate.A 503 bp tandem repeated nucleotide fragment was found inserted at the 3′end of the genome,which has 96%homology with part end sequences of the strains from outside of China.This variant does not contain any known marker genes,indicating that natural variants of ASFV are occurring in China,and this may be related to the subacute epidemic of ASF in our country.
