摘    要:目的]建立和优化芸香科药用植物的品种的ISSR反应体系,为芸香科药用植物的资源评价、品种鉴定提供依据。方法]以象头山芸香科植物柚、山油柑、3个品种为研究对象,从DNA提取、退火温度、循环次数、引物筛选等方面研究ISSR反应及优化条件。结果]选用的20个引物中有3种引物扩增效果较好,以柚为代表的优化结果表明,引物826对芸香科植物扩增的最适退火温度为52.0℃,最佳循环次数为35个循环。结论]ISSR适合于芸香科药用植物的品种的鉴定。

Construction and Optimization of ISSR Reaction System of Medicinal Rutaceae Plants
Abstract:Objective] The research aimed to study the construction and optimization of ISSR reaction system of medicinal Rutaceae plant,and provide references for resources evaluation and cultivar identification of Rutaceae plants.Method] Three varieties of Rutaceae plants including C.grandis(L.)Osbeck,Acronychia pedunculata(L.)Miq.,Z.avicennae(Lam.)DC were used as material for study.The ISSR reaction system included the genomic DNA extraction,amplification,cycle time,primers screening.Result] In the Identification of ISSR molecular markers,3 primers in 20 of being chose had better amplification effect.Optimization results representative of C.grandis(L.)Osbeck showed that primers 826 for the optimum temperature of Rutaceae plants amplification was 52.0 ℃,and the best cycle time was 35 cycles.Conclusion] ISSR reaction system can used to identify the cultivar of Rutaceae plants.
