鸳鸯茉莉不同外植体诱导愈伤组织研究初探

以鸳鸯茉莉花瓣、叶片、茎段为外植体,MS为基本培养基,探索外植体取材季节和部位对污染的影响;分析植物生长调节剂的种类、浓度对不同外植体诱导愈伤组织的影响和最适组合。结果表明:在34、月取材比5、6月好,12月取材效果较差;通过花瓣能成功诱导出健康的愈伤组织,但诱导率较低(27.1%),平均出愈时间为23~25 d,3种外植体中以嫩叶诱导愈伤组织的效果最好(87.1%),平均出愈时间最短,为18~20 d;茎段愈伤组织诱导率其次(68.1%),平均出愈时间为21~23 d;2,4-D6、-BA对以花瓣为外植体诱导愈伤组织的影响达显著水平,花瓣诱导愈伤组织的最适培养基为MS+6-BA 1.0 mg/L+NAA 1.5 mg/L+2,4-D 1.0 mg/L,2,4-D对叶片诱导愈伤组织的影响达显著水平,叶片诱导愈伤组织的最适培养基为MS+6-BA 2.0 mg/L+NAA 1.0 mg/L+2,4-D 1.0 mg/L,2,4-D、6-B对茎段诱导愈伤组织的影响达到极显著水平;MS+6-BA 3.0 mg/L+KT 3.0 mg/L+NAA 1.0 mg/L+2,4-D 1.0 mg/L最适合茎段诱导愈伤组织。
