摘    要:建立了分离7种酚类物质的高效液相色谱条件,对低温贮藏过程中兰州百合鳞茎不同部位的酚类物质成分和含量进行了测定。采用Waters Nova-Pak■C18色谱柱(3.9 mmi.d.×150 mm,4μm),柱温35℃,流动相为乙腈—1%(V/V)乙酸水溶液(15∶85),流速1.0 mL/min,紫外检测波长280 nm,进样量15μL,可使7种酚类物质在20 min内达到完全分离。鳞片、顶芽和鳞茎盘内均可检测到对羟基苯甲酸、阿魏酸、香豆酸、水杨酸和香豆素。其中对羟基苯甲酸的含量显著高于其它酚类物质的含量。不同贮藏时期、不同鳞茎部位酚类物质的种类和含量存在差异。

Abstract:To determine phenolics in different parts of Lilium davidii var.unicolor during bulb storage at low temperature,the conditions of high performance liquid chromatography(HPLC) were established with Waters Nova-Pak■C18 column(3.9 mm i.d.×150 mm,4 μm),and the column temperature was 35℃.Acetonitrile-1%(V/V) acetic acid(15∶85) was used as the mobile phase with the flow rate of 1.0 mL/min.The detective wavelength was 280 nm and the injection amount was 15 μL.Phenolics can be separated completely within 20 min.The results indicated that 4-hydroxybenzoic acid,ferulic acid,m-coumaric acid,salicylic acid and coumarin existed in scales,buds and basal plates.The content of 4-hydroxybenzoic acid was much higher than other phenolics.The kinds and contents of phenolics were different in various parts of bulb and storage stages.
