摘    要:目的]研究高原地区粗柄羊肚菌生长的生态条件及适宜的菌种分离方法。方法]将野外调查和实验室组织分离方法相结合,以污染率和菌丝长势为指标,比较不同分离方法的优劣。结果]子实体发生温度约为15℃,土壤呈碱性;土壤中菌丝长势纤细,未有肉眼可见菌索、菌根等结构,采用组织分离和基质分离的方法成功率低,污染率高。结论]粗柄羊肚菌子实体发生的限制因素是温度,菌丝特化的子囊菌宜采用孢子分离的方法获取菌种。

Ecological Conditions of Morchellaceae crassipes and Optimization of Strain Separation

Abstract:Objective] To study the ecological growth conditions and strain separation method of Morchellaceae crassipes.Method] Through the combined method of field investigation and laboratory separation,the advantages and disadvantages of several separation methods were compared according to the contamination rate and mycelium growth vigor.Result] Fruitbody occurred when the temperature rose up to 15 ℃,the soil became alkaline.As the hypha was fine in the soil and no rhizomorph or fungi root was observed by naked eyes,strain separation could hardly be achieved due to high contamination from soil matrix or fruitbody tissue.Conclusion] The fruitbody formation of Morchellaceae crassipes is constrained by temperature,and spore separation is suitable for obtaining mycelium from spores of ascomycetes.
