摘 要: | [目的]研究牛大力组织培养与快速繁殖技术。[方法]以牛大力种子为外植体,采用以芽繁芽途径,比较3种不同种源牛大力种子无菌发芽率、继代增殖、生根培养的表现差异。[结果]以MS+6-BA 0.4 mg/L+NAA 0.2 mg/L为诱导培养基,种子无菌发芽率分别为98.23%、92.13%和95.01%;以改良MS+6-BA 0.6 mg/L+IBA 0.3 mg/L+KT 0.2 mg/L为继代增殖培养基,添加半胱氨酸8 mg/L,FeSO_4·7H_2O加至MS用量的1.2倍,增殖系数分别达5.57、3.20、5.30;以1/2MS+NAA 1.0 mg/L+IBA 1.5 mg/L、1/2MS+NAA 1.0 mg/L+IBA 3.0 mg/L、1/2MS+NAA 1.5 mg/L+IBA 1.0 mg/L为生根培养基,生根率分别达78.00%、67.33%、70.33%;生根苗移栽在V黄心土∶V细河沙∶V泥炭土=8∶1∶1基质中培育60 d后,成活率分别达94.00%、85.43%和89.97%。采用控根容器培育大苗,可实现育苗与种植同步。[结论]该研究为牛大力种苗快速繁殖及产业化生产提供科学依据。

Abstract: | Objective] The aim was to research the tissue culture and rapid propagation of Millettia speciosa.[ Method] Using M.speciosa seeds as explant, the seeds germination rate, the proliferation coefficient, the rooting rate of M.speciosa from three provenances were compared.[Re-sult] The results showed that the seeds germination rate was 98.23%, 92.13% and 95.01% respectively under sterile condition in medium of MS+6-BA 0.4 mg/L+NAA 0.2 mg/L, the proliferation coefficient was 5.57, 3.20 and 5.30 respectively in medium of modified MS+6-BA 0.6 mg/L +IBA 0.3 mg/L+KT 0.2 mg/L, added with cysteine 8 mg/L and 1.2 times of FeSO4 · 7H2 O to MS, the rooting rate was 78.00%, 67.33% and 70.33% respectively in medium of 1/2MS+NAA 1.0 mg/L+IBA 1.5 mg/L,1/2MS+NAA 1.0 mg/L+IBA 3.0 mg/L and 1/2MS+NAA 1.5 mg/L+IBA 1.0 mg/L, and the transplanting survival rate was 94.00%, 85.43%and 89.97%respectively on yellow sub-soil, fine sand and peat soil at the rate of 8∶1∶1.The root controlling container seedlings cultivation, breeding and planting can achieve synchro-niaztion.[Conclusion] The research can provide the scientific basis for seedling rapid propagation and industrial production of M.speciosa. |
