摘    要:目的]比较金太阳杏花粉量及花粉活力的测定方法。方法]采摘金太阳杏各单株已达气球期、饱满、正常、未开裂的花蕾,收集花粉,通过染色法、离体萌发法测定花粉活力,研究不同浓度的蔗糖、硼酸对花粉萌发花粉管的影响,并测定每枚花药花粉量。结果]通过2,3,5-氯化三苯基四氮唑(TTC)染色法测定花粉活力为11.50%。对于离体萌发法,当培养温度为25℃、培养基p H为6.5、蔗糖浓度为10%、硼酸浓度为25μg/m L时花粉萌发花粉管效果最好,此时花粉活力为27.00%。金太阳杏花粉量测定用单枚花药花粉量来衡量,测得单枚花药花粉量平均为1 303。结论]试验结果为提高育种效率提供了理论依据。

Detection of Pollen Quantity and Vitality of Golden Sun Apricot
Abstract:Objective] To compare the determination methods for pollen quantity and vitality of Golden Sun Apricot.Method] Pollen from full normal un-blossomed buds was picked from wild peach at the balloon stage .And the pollen viability was determined by the means of staining method and in vitro germination method .Effects of different concentrations of saccharose and boric acid on the pollen tube germination were re-searched.And the pollen amount of the single semet was determined.Result] The pollen viability was determined by TTC staining method was 11.50%.Under 25 ℃ temperature, medium pH 6.5, 10% saccharose concentration, and 25 μg/mL boric acid concentration, the effects of sprouting reached the highest (27.00% pollen viability).The pollen amount of Golden Sun Apricot was measured by determining the pollen a-mount of the single semet, and the pollen amount of the single semet was 1 303.Conclusion] The test results provide theoretical foundation for enhancing breeding efficiency.
