银杏愈伤组织细胞中总黄酮提取工艺的比较

摘    要:为探索从药用植物细胞中提取黄酮类活性物质,用简单、易工业实现的低成本工艺获得较大的总黄酮提取率,为工业生产提供理论依据和实践指导,以银杏愈伤组织为材料提取总黄酮并测定含量。利用乙醇提取、超声波提取和水提取3种方法对银杏愈伤组织中黄酮类化合物进行提取,以络合分光光度法测定的黄酮得率为衡量指标,得到各自最佳的提取条件。结果表明:利用乙醇法提取愈伤组织中总黄酮提取最佳条件是:80%的乙醇作为提取剂,料液比为1∶7(m∶v),于60℃提取3次,总黄酮得率为3.67%;超声波法提取最佳条件为60%的乙醇作为提取剂,料液比为1∶34(m∶v),提取65 min总黄酮,得率为4.57%;水提法的最佳条件是:水温90℃,料液比为1∶30(m∶v),提取3.5 h,总黄酮得率为2.95%。3种优化后的提取方法黄酮得率依次为:超声法>乙醇法>水提法。
