摘    要:作者通过对融合方式、融合场强、体细胞与透明带切口位置及重构后是否修复等条件对北山羊—山羊体细胞核移植重构胚融合率的影响进行了研究。结果表明,不同融合方式能显著影响融合率(67.57%和42.15%,P<0.05);场强在1.3~1.9 kV/cm范围内对融合率和死亡率都没有影响;体细胞在透明带的切口处对融合率影响不显著,但融合过程中死亡率高于对照组(8.25%和2.3%,P>0.05);重构后立即融合与修复2 h后再融合对融合率没有影响(67.29%和68.33%,P>0.05)。

Abstract:This paper investigated the effects of different conditions on the fusing rate of Capra ibex-goat somatic cell nuclear transfer reconstructed embryo, including fusion means, fusion strength, incision site between Somatic and zona pellucida as well as it whether repaired after reconstructed and so on. The results showed that different fusion means had significant effects on the fusing rate(67.57% and 42.15%,P<0.05);it had no effect on the fusing rate and death rate when the fusing strength IS between 1.3 to 1.9 kV/cm; the incision site between Somatic and zona pellucida had no significant influence on fusing rate, but the oocyte’ dead rate was higher than the control’s during fusion (8.25% and 2.3%,P>0.05 ); it had no effect on fusing rate between immediately fusion after reconstructed and fusion after repairing 2 h(67.29% and 68.33%,P>0.05).
