绞股蓝皂苷最佳提取条件的研究

摘    要:目的]确定绞股蓝皂苷的最佳提取条件。方法]制备标准曲线,用香草醛—高氯酸比色法测定绞股蓝皂苷的含量,通过正交试验研究浸提次数、浸提温度、乙醇浓度、料液比不同水平对绞股蓝皂苷提取率的影响。结果]浸提次数、浸提温度、乙醇浓度、料液比对绞股蓝皂苷提取率的影响依次减小,浸提温度和浸提次数对提取绞股蓝皂苷的影响明显大于乙醇浓度和料液比。直观性分析表明,绞股蓝皂苷的最佳提取工艺为:70%乙醇溶液,料液比1∶12,在40℃下浸提3次,每次2 d。F检验表明,在3个乙醇浓度水平和3个料液比水平内提取绞股蓝皂苷的效果无显著差异,而温度和浸提次数各水平间的差异显著。结论]在绞股蓝皂苷的提取过程中,不同提取条件对绞股蓝皂苷的提取率有很大的影响。

Abstract:Objective] The purpose was to confirm the optimum condition of extracting gypenosides.Method] The standard curve was prepared and vanillin-perchloride acid colorimetry was used to determine gypenosides content.The influences of extraction times,extraction temperature,ethanol concentration and material-liquid ratio with different levels on the extraction rate of gypenosides were studied through orthogonal test.Result] The influences of extraction times,extraction temperature,ethanol concentration and material-liquid ratio on the extraction rate of gypenosides minished in order.The influences of extraction temperature and extraction times on gypenosides were obviously higher than that of ethanol concentration and material-liquid ratio.The intuitive analysis showed that the optimum extraction technology was with 70% ethanol and material-liquid ratio of 1∶12,extracting for 3 times and 2 d once at 40 ℃.F test showed that the effects of extracting gypenosides had no significant difference among 3 ethanol concentration levels and 3 material-liquid ratio levels,but the differences among various levels of temperature and extraction times were significant.Conclusion] In the extraction course of gypenosides,the difference of extracting conditions had very big influence on the extraction rate of gypenosides.
