产不同色素古尼拟青霉菌株功效成分分析

摘    要:为比较产不同色素古尼拟青霉菌株菌丝体功效成分的差异,本研究对产不同色素古尼拟青霉菌株及对应古尼虫草样品进行试验分析。观察其菌落形态和菌落颜色;用紫外分光光度法测定多糖和D-甘露醇含量;用HPLC法测定虫草素、腺苷、肌苷、胸苷、鸟苷、尿苷6种核苷类成分含量。结果显示,菌落背面绿色的菌株pgjt03,腺苷含量是古尼虫草样品YHCGGB0145的6倍,且虫草素、胸苷、鸟苷、尿苷含量也均达到最高,与它对应的古尼虫草样品YH—CGJT0148虫草素、腺苷、鸟苷、尿苷含量均高于其它古尼虫草;而菌落背面浅灰色的菌株pNt01,核苷类成分含量均低于其他菌株。说明古尼拟青霉产色素与功效成分之间有一定的相关性,古尼拟青霉所产色素颜色越深,功效成分的含量相对越高。通过该研究指出,菌落颜色可作为筛选古尼拟青霉菌株高产功效成分的标志之一,且菌落背面绿色的菌株pgit03,性状优良,生长较快,可作为潜在的生产菌株。

Abstract:In order to compare the effective components with the different pigment strains of Paecilomyces gunnii, the colonial morphologies and colors of different pigment strains were observed. The contents of cordycepic polysaccharide and D - mannitol were determined by ultraviolet spectroscopy. The cordycepin, adenosine, thymidine, vernine, inosine and uridine were deter- mined by HPLC. The analytical results showed that the strain pgjt03 with colony green back side had the highest contents of cordycepin, thymidine, vernine and uridine in all of strains, it was 6 times than YHCGGB0145 in adenosine, and the counter- part sample YHCGJT0148 of Cordyceps gunnii had the higher contents of cordycepin, cordyceps adenosine, vernine and uridine than ones of the rest samples of C. gunnii. Reversely, the strain pgjt01 with colony hoary back side had the lower contents of nu clcosides than ones of the rest strains. It was illustrated that there were some relativities between the pigment strains of P. gunnii and the effective components, and darker pigment strains of P. gunnii contained higher the effective components. Through the study, it was implied that the colonial color of strains might be as a maker to select higher effective components of P. gunnii, and the strain pgjt03 with colony green back side could be potential production strain, for having superior traits and fast growth.
