摘    要:本研究利用生物信息学并结合分子生物学实验对日本血吸虫细胞因子诱导凋亡因子CIAPIN1(cytokine induced apoptosis inhibitor 1)基因进行了初步研究。研究表明日本血吸虫存在CIAPIN1同源基因,将其命名为SjCIAPIN1,SjCIAPIN1基因已知cDNA长1143 bp,包括完整的CDS,编码276个氨基酸,预测分子量为30.55863 kDa。通过原核表达系统对该蛋白进行了重组表达和纯化,并制备了多克隆抗体制备。Western blot分析表明,本研究制备的多克隆抗体能与SjCIAPIN1重组蛋白特异识别。

Abstract:In the present study,bioinformatics and molecular techniques were used to preliminarily study cytokine induced apoptosis inhibitor 1(CIAPIN1) in Schistosoma japonicum.Bioinformatical analysis revealed that there was a CIAPIN1 analogous in Schistosoma japonicum,named SjCIAPIN1.The cDNA of SjCIAPIN1 is 1143bp in length,encoding 276 amino acid residues.The cDNA fragment containing the open reading frame of SjCIAPIN1 was cloned into a pET28a(+) vector to construct the recombinant plasmid for protein expression.The recombinant protein was purified by affinity purification and then injected to New Zealand white rabbits for preparing the polyclonal antibodies against SjCIAPIN1.Western blot analysis indicated that the polyclonal antibodies were able to specifically recognize the recombinant SjCIAPIN1 protein.
