茶梅花瓣总RNA提取方法的比较和分析

摘    要:总RNA的纯度和完整性对植物分子生物学实验至关重要,为了探索更适合茶梅花瓣总RNA的提取方法,本研究分别利用CTAB多级沉淀法、改良的异硫氰酸胍法、改良的热硼酸法、Trizol法和EASY spin植物RNA提取试剂盒法等5种方法提取茶梅花瓣总RNA,并进行了对比分析。结果发现EASY spin植物RNA提取试剂盒法是最为简单、快速、高效的方法,该方法得到的RNA条带在琼脂糖凝胶上有清晰的28S、18S和5S 3条带,且超微量分光光度计测量的A260/280、A260/230均在允许范围内,表明该方法得到的RNA完整、质量高。CTAB多级沉淀法得到的RNA质量次之。本研究为今后有效开展茶梅分子生物学研究提供技术便利,并为其他花卉植物RNA的提取提供必要的参考。

Abstract:The total RNA from petals of Camellia sasanqua was extracted by the methods of CTAB-many-level-precipitation method, improved isothiocyanate method, improved hot-borate method, Trizol method and EASY spin plant RNA extraction Kit method, respectively, and the five methods were contrasted and analyzed. The results showed that the method of EASY spin plant RNA extraction Kit was the simplest, the quickest and high efficient, and CTAB-many-level-precipitation method took the second place. The study provided convenient and effective technical supports for the research on molecular biology of Camellia sasanqua, and also provided necessary reference for the RNA extraction of other flowers.
