| 摘 要： | 人源性胰岛素样生长因子1(hIGF-1)是一类在许多人类疾病治疗中都具有重要作用的细胞因子.本研究成功构建了以水稻种子高效表达的谷蛋白Gti基因5.3kb启动子引导hIGF-1基因(higf-1)的表达载体:pCAMBIA 1301-5.3kbGt1-higf-1-nos.在此基础上,将构建好的表达载体,通过根癌农杆菌介导法转化水稻,获得转基因植株.通过对转基因水稻植株叶片总DNA的PCR检测,初步确定表达载体的T-DNA区段已经成功整合入水稻基因组中.目的基因RT-PCR检测结果显示,higf-1在转基因水稻种子中能够正常转录.开展本试验为利用水稻及其他单子叶植物表达IGF-1研究奠定了基础.
