Inhibiting effect of doxycycline on intimal proliferation after percutaneous transluminal angioplasty in rabbits

Abstract:Objective To investigate the effect of doxycycline on intimal hyperplasia and its action mechanism after percutaneous transluminal angioplasty (PTA) in rabbits. Methods New Zealand white rabbits were fed by high-fat diet to construct the artherosclerosis models, and then randomly divided into sham operation, simple PTA, and three-dose doxycycline (10, 30, and 50 mg · kg^-1·d^-1) groups, each of 6 animals. Doxycycline groups were treated with doxycycline solution two days preoperation, while sham operation and simple PTA groups with normal saline. All rabbits were sacrificed after 8 weeks, and their common jugular arteries were removed. Plasma level of endothelin-1 (ET-1), intimal expression of proliferating cell nuclear antigen (PCNA), and histomorphometry were detected by ELISA, immunohistochemistry, and HE stain, respectively. Results Compared with sham operation group, there were significant intimal hyperplasia and upregulation of ET-1 level and intimal PCNA expression in simple PTA group (P〈0.01). However, three-dose doxycycline treatment reduced intimal hyperplasia, ET-1 content and intimal PCNA expression compared with simple PTA group (P〈0.01). Conclusion Doxycycline can effectively inhibit intimal proliferation after angioplasty, which may be due to downregulation of plasma ET-1 level and intimal PCNA expression.
