马缨杜鹃下胚轴组织培养及全息现象的研究

摘    要:以马缨杜鹃下胚轴为外植体,用L9(34)正交实验法研究基本培养基、NAA、2-ip对芽器官诱导的影响。以马缨杜鹃下胚轴切断为材料,研究全息现象。结果表明马缨杜鹃下胚轴诱导芽的最佳培养配方为1/4 MS+NAA0.75 mg/L+2-ip2.0 mg/L。马缨杜鹃切段在离体培养时存在全息现象,其不定芽发生的部位及数量、频率的梯度变化都遵循生物全息律,全息律的呈现在一定程度上受外源激素和切断位置的影响。

Abstract:Using Rhododendron delavayi Franch hypocotyl as explant,through the L_9(3~4) orthogonal experiment,a study was conducted to study the effect of tissue culture on organs sprouting by basic culture medium、NAA、2-ip.Moreover,researched bioholographic phenomenon by hypocotyl cutting tissue culture.Results showed that the optimal combination of bud differentiation Was 1/4 MS+0.75 ms/L NAA+2-ip 2.0 mg/L.It Was found that in vitro culture of hypocotyl segments showed holographic phenomenon;the position and the quantity、the frequency gradient of sprouting inducing all conformed to the bioholographic law,the hormone and the position of hypocotyl could effect the bioholographic law to a certain extent.
