摘    要:
大豆成熟子叶外植体,培养在附加2.0mg/LBA的MS培养基上,诱导不定芽的发生,培养10天后分化出不定芽,30天后长出无根小苗。在不定芽发生过程中,经SDS-PAGE分析表明,19kD、36kD、48kD和72kD4条蛋白质条带在培养第5天时消失,培养第4天后出现25kD和27kD新条带,第5天又出现32kD和33.8kD新条带,同时,55kD蛋白质含量明显增加(条带由浅变深),82kD和低于1

Physiological and Biochemical Characteristics of Adventitious Bud Formation in Soybean Mature-Cotyledon In vitro Culture

Abstract:
Maturecotyledon explants of soybean were cultured on MS medium with 2.0mg/L BA. The adventitious buds were differentiated after 10 days of culture and rootless plantlet were obtained after 30 days of culture.It was shown that by SDS-PAGE analysis, the bands of four proteins with molecular weights of 19kD, 36kD, 48kD and 72kD respectively disappeared at the fifth day of culture, the new bands of proteins with molecular weights of 25kD and 27kD appeared at the fourth day of culture, and those of proteins with 32kD and 33.8kD appeared at the fifth day of culture. The content of protein with molecular weight of 55kD increased obviously after 5 days of culture and the protein with molecular wight of 82kD and those with molecular weights below 16kD were stable during the culture.It was observed that there were two new bands of isoperoxidase appearing at the negative pole after 2 days of culture and two new bands appearing at the positive pole on the fourth day and fifth day of culture, respectively. The ctent of soluable proteins decreased with an increase in culture time. The activity of peroxidase had two peaks at the fifth day and seventh day of culture and the content of endogenous IAA had two lowest of values at the second day and fifth day, respectively. As adventitious buds initiated, the content of soluable proteins increased and the activity of peroxidase decreaseed; the contents of endogenous IAA and ZR reached a highest level, and the four new bands of isoperoxidase appearing at the positive and negative poles disappeared.
