摘    要:为完善用于蛋白质组分析的双向电泳体系,以"久保"桃中果皮为试材,比较pH 5.0~8.0与pH 3.0~10.0线性范围的固相pH梯度(immobilized pH gradients,IPG)胶条对蛋白质双向电泳的影响。结果表明:采用pH 5.0~8.0的17 cm胶条,90μg蛋白上样量,等电聚焦(IEF)电泳后,进行十二烷基磺酸钠聚丙烯酰胺凝胶电泳(SDS-PAGE),银染色得出的双向电泳图有蛋白点(1 318±125)个,图谱背景清晰效果较好,适用于桃中果皮蛋白质组分析。

Abstract:Peach(Prunus persica L.cv.Okuba) mesocarp was adopted in this experiment to consummate the protocol of two-dimensional electrophoresis for proteomic analysis and compare the effect of two-dimensional electrophoresis(2-DE) maps that were involved with immobilized pH gradients(IPG) strips with the linear range of pH5.0~8.0 and pH3.0~10.0.Approximately 90 μg of protein was loaded on 17 cm IPG strip which extension of hydrogen ion exponent was 5.0~8.0,after isoelectric focusing(IEF) completed sodium dodecyl sulfatepolyacrylamide gel electrophoresis(SDS-PAGE) was processed.The two-dimensional gel stained by silver contained 1318±125 protein spots and had a clear background on the map.The results demonstrated that this protocol was suitable for proteomic analysis of peach mesocarp.
