晚熟桃微繁技术的研究

摘    要:采用正交试验法对晚熟和中晚熟两个鲜食桃品种试管苗丛生芽诱导增殖和生根培养基进行了筛选,并对其组培微繁体系进行了研究。结果表明:MS+1.0 mg/L 6-BA+0.5 mg/L GA3+2.0 mg/L IBA培养基丛生芽诱导率最高,可达93.8%;以1/2 MS附加100 mg/L IBA培养液浸蘸丛生芽基部,再在16℃室温下暗培养8 d,生根率可达 85%,但浸蘸时间的长短与诱导生根效果关系不明显。通过逐渐降低湿度和分步炼苗,生根试管苗移栽成活率可达 80%以上。

Abstract:Experiment of bud induction of fresh meal peach was conducted by using the orthogonal experiment design. The micropropagation of late-maturing Prunus persica was studied. The results showed that the suitable medium for the bud induction from shoot tips was MS+1. 0 mg/L 6-BA+ 0. 5 mg/L GA3 +2. 0 mg/L IBA,the rate of the bud induction could reach 93. 8 %. 1/2 MS with soaking the root in 100 mg/L IBA and 16 ℃ dark treatment for 8 days was the best for inducing root,the rooting rate was 85 %. There was no significant relation between soaking time and rooting rate. Through reducing humidity gradually and three-step acclimatization procedure,the survival rate could be over 80 %.
