淋巴细胞性J亚群禽白血病的诊断及其病理组织学观察

摘    要:对2例疑似感染J亚群禽白血病病毒(J-Subtype Avian leukosis virus,ALV-J)的病禽进行临床观察和病理解剖,结果发现肝脏和脾脏均肿大,肝脏、心脏、肺脏、腺胃有肿瘤样结节;设计1对引物针对ALV-J的gp85基因,对2例病死鸡的肝脏组织进行PCR检测,结果 2份组织样品均扩增出与预想结果一致的924 bp的ALV-J特异片段;取病死禽的心、肝、脾脏等组织制做石蜡切片,HE染色,进行病理组织学观察,结果发现肝组织有大量成淋巴细胞和淋巴样瘤细胞,呈灶状聚集;脾组织结构松散,白髓和红髓界限不清,脾小体消失,间隙有大量淋巴样瘤细胞;心包脂肪、心肌间、心脏血管中大量淋巴样瘤细胞;腺胃肌层、腺胃乳头间隙有淋巴样瘤细胞,1病例腺胃乳头可见出血;肺泡隔及肺泡腔中有大量淋巴样瘤细胞填充等病理变化。试验结果证实,2例土鸡均为淋巴细胞性J亚群禽白血病。

Abstract:Two cases of sick chickens, which?were?suspect to?be?infected?with J-Subtype?Avian?leukosis virus (ALV-J), were?diagnosed?by?clinical?observation?and?pathological?anatomy. The?results?indicated that swollen and tumor nodules were observed in liver, heart, spleen and lung. Based?on?the published gp85?sequence?of?ALV-J, a pair?of?PCR primers?was?designed?to amplify?the?nucleic?acids?of?ALV-J. The?result?found?that?a 924 bp specific?band?that agrees?with?the?expected size of?ALV-J?gp85?was?amplified?from?two??liver?tissues. The?organs in diseased chickins,?such?as?liver,?heart,?spleen,?glandular?stomach,?and lung?were?analyzed after soaking in 10% formaldehyde solution,?paraffin?embedding, and HE?dyeing.The?results?showed?that large areas of the pericardial fat, myocardium, heart blood vessels and spleen, liver, lung, glandular stomach were infiltrated by lymphocytes and lymphoid cells. Glandular stomach nipple?showed congestion and hemorrhage in one case. Those results confirmed the lymphocytic subgroup J-subtype avian leucosis in two local chickens in Anhui area.
