摘    要:[目的]研究3种不同配方青蒿琥酯(Artesunate,AS)纳米乳的急性毒性,筛选出安全性最好的配方。[方法]采用小鼠灌胃给药,观察14 d内小鼠的活动情况及死亡情况,记录小鼠的死亡数。用改良寇氏法测定各配方AS纳米乳的半数致死量(LD50)及可信限。[结果]AS1、AS2纳米乳的LD50分别为7.76和7.56 g/kg;可信限为6.90~8.68和6.42~8.87。AS3纳米乳的LD0为15 g/kg。[结论]在国内首次成功制备的3种不同配方AS纳米乳,均属于实际无毒范围,AS3纳米乳较之AS1、AS2更安全。

Study on the Acute Toxicity of Artesunate Nano-emulsion in Mice by Oral Administration
Abstract:[Objective] The research aimed to study the acute toxicity of 3 kinds of artesunate with different formula and screen out the formula with the best safety.[Method] The drugs were infused into stomach of mice to observe the activity situation and death situation of mice within 14 days.The number of dead mice was recorded.The median lethal dose(LD50) and confidence limit of AS nano-emulsion with different formula were determined by using improved Karber method.[Result] LD50 of AS1 and AS2 nano-emulsion were 7.76 and 7.56 g/kg resp.and the confidence limit of the two were 6.90-8.68 and 6.42-8.87 resp.LD50 of AS3 nanoemulsion was 15 g/kg.[Conclusion] 3 kinds of nano-emulsion with different formula prepared successfully in China for the time all had no actual toxicity.AS3 nano-emulsion was safer than AS1 and AS2.
